SPIFAN ERP-Approved First Action Methods to Be Published in March/April 2012 Issue of J. AOAC Int.

The first collection of AOAC Stakeholder Panel on Infant Formula and Adult Nutritionals (SPIFAN) methods will be published in the March/April 2012 issue of the Journal of AOAC INTERNATIONAL and are available on the Official Methods of AnalysisSM Online (www.eoma.aoac.org). The methods were the first to be approved by an expert review panel (ERP) based on evaluation against standard method performance requirements (SMPRs) developed by SPIFAN.

"The new First Action Methods in the Journal come from the infant formula community and are state-of-the art,” said Darryl Sullivan of Covance Laboratories and chair of SPIFAN. “This is going to bring infant formula testing forward 25 years in technology. With HPLC and MS/MS techniques, the methods are highly specific. SPIFAN established stringent SMPRs, requiring high performance from the methods. We are confident in these methods, which operate well within the Horwitz curve."

On June 29, 2011, as part of the “Standards Development and International Harmonization: AOAC INTERNATIONAL Mid-Year Meeting,” the inaugural ERP charged with granting First Action status evaluated methods for the first set of priority nutrients against SMPRs. All candidate methods for vitamins A, B12, and D, and folic acid met SMPRs (appropriateness for the intended use, clarity of the method description, recovery, analytical range, and limits of detection and quantitation) and, consequently, were approved First Action. The first set of methods to be published includes:

* 2011.07 Vitamin A in Infant Formula and Adult Nutritionals by UPLC-UV

* 2011.08 Vitamin B12 in Infant Formula and Adult/Pediatric Nutritional Formula by Liquid Chromatography/UV Detection with Immunoaffinity Extraction [Campos-Giménez, E., Fontannaz, P., Trisconi, M-J., Kilinc, T., Gimenez, C., & Andrieux, P. (2008) J. AOAC Int. 91, 786–793]

* 2011.10 Vitamin B12 in Infant Formula and Adult/Pediatric Nutritional Formula by HPLC

* 2011.11 Vitamin D in Infant Formula and Adult/Pediatric Nutritional Formula by Ultra-High-Performance Liquid Chromatography-Tandem Mass Spectrometry

* 2011.15 Vitamin A (Retinol) in Infant Formula and Adult Nutritionals by Liquid Chromatography [previously approved as AOAC Official MethodSM 2001.13 (for foods), J. AOAC Int. 85, 424(2002)]

* 2011.16 Vitamin B12 in Infant Formula and Adult/Pediatric Nutritional Formula by Surface Plasmon Resonance [Indyk, H.E., Persson, B.S., Caselunghe, M.C.B., Moberg, A., Filonzi, E.L., & Woollard, D.C. (2002) J. AOAC Int. 85, 72–81]

To date, ERPs have adopted 15 First Action Methods for the SPIFAN initiative. All methods have been codified and are currently being drafted into AOAC style for publication.

"Approval and publication of these methods makes them available to laboratories, so that they can start using them and generating additional information on the methods’ performance,” said Sullivan. “AOAC can make use of this valuable information to determine if the methods need a change in status or additional information."
