AOAC INTERNATIONAL has received a proposal to modify AOAC 995.03 Capsaicinoids in Capsicums and Their Extractives.

AOAC has issued a Call for Comments for the proposed modification of AOAC 995.03, Capsaicinoids in Capsicums and Their Extractives. AOAC is collecting comments on the proposal, including the rationale and proposed modification. Both the rationale to modify the method and the proposed modification are as follows:
Rationale: In the first paragraph of the method, a note indicates “1 mg total capsaicinoids/g ca 15 SHU”, thus providing an option for the analyst to ‘calculate’ the sample capsaicinoids content. There has been experience indicating that the “15 SHU” approximation may the source of significant discrepancies (around 8% relative) when comparing capsaicinoid contents calculated directly from chromatographic data vs. derived from SHU values. It is believed that the response factor for capsaicin by UV detection, as published in the method, is erroneous.
Proposed Modification: To address the error in response factor identified for capsaicin by UV detection, it is suggested to independently verify the response factors for all three capsaicinoids in the method using commercially available reference materials, both by UV and fluorescence detection
AOAC invites all users and stakeholders that used or incorporate AOAC 995.03 to provide comments on the proposed modification for a minimum of 30 – 60 days. All comments will be reviewed and reconciled.
